As noted by Locke et al (PMID: 21084631), Kaplan's in vitro dataset was derived from an experiment using a 0.4:1 mass ratio of histone to DNA. A more natural ratio of 1:1, corresponding to actual levels in vivo, was used by a previous report by Zhang et al (PMID: 19620965), also concerning eukaryotic chromatin studied in vitro. The correlation between the two databases was rather low (r = 0.69). Because the Zhang publication appeared about ten weeks prior to the submission of Kaplan's manuscript to Nature, Kaplan could have noted this discrepancy for consideration by their readers. |
